Sample Pooling for SARS-CoV-2

Sample pooling is a testing approach where we pool multiple samples into one sample for qPCR testing. It is a COVID-19 screening strategy that has been approved for FDA Emergency Use Authorizations(EUA) applications. Sample pooling is also approved by the Centre of Health Protection (Hong Kong). COVID-19 RT-PCR testing institutions that are recognized by Hong Kong Government can provide sample pooling testing services to the public given they pass the External Quality Assessment Programme (EQAP).

HKLife’s Unique Sample Pooling Protocol

The sample pooling approach adopted by other testing laboratories suffers from reduced sensitivity for pooled samples. This is due to the introduction of dilution when multiple samples were pooled together for extraction. HKLife developed our own unique sample pooling protocol which requires no dilution of samples . Samples using our unique pooling protocol will retain the same level of sensitivity as individual testing, yet receives all the benefits from sample pooling.

One swab is taken from each person and placed in separate sampling tubes containing 1 mL specimen media. Take 200 μL from each of the 5 tubes and mix, and use the 1000 μL pooled sample for nucleic acid detection. If the pooled sample is positive, use the remaining specimen media for individual testing.

Key Benefits
Enhance throughput / Reduce cost
Test the same amount of samples with fewer tests
Test the same amount of samples with fewer testing supplies
Test the same amount of samples with fewer manpower
Test more samples with less time
Reduced reporting time for your clients

Training Course

As to popularize the usage of sample pooling for large sample volume, we would like to invite you to join our “Sample Pooling Method for Molecular Analysis” training course.

In this course, you can:

  1. Understand the principle of sample pooling, DNA extraction and real time qPCR analysis
  2. Understand the advantage of the sample pooling method and precaution
  3. Establish the experimental design for the pooling verification
  4. Identify the limitation of the sample pooling method
  5. Select the appropriate sampling method for the DNA analysis
Course Outline
1. Importance of Sample Pooling
2. Introduction of Common Sample Pooling Approach
3. Working Principle of Repeated Column Binding
4. Laboratory Safety and Precautions

FAQs

With new SARS-CoV-2 variants emerging at such a high frequency, HKLife understands the importance in being able to detect all the variants because any non-detected case can lead to further transmission and outbreaks. That is why we make every effort to continuously monitor the emergence of new variants and ensure that the performance of our products are not compromised by any new mutations. HKLife proudly announce our C/Tech SARS-CoV-2 products are able to detect all variants to date including the Delta and Omicron variants.

All HKLife test kits are manufactured in Hong Kong Science Park under the standard of ISO13485.

We have conducted a series of comparative studies to investigate on the sensitivity of HKLife’s range of products. The information can be found here. Our product is also being used by HK Government recognized COVID-19 testing institutions for SARS-CoV-2 testing and has passed the QAP scheme for all collaborating laboratories (Individual testing and 3 in 1 pooled testing).

Sample pooling refers to a testing approach designed to efficiently screen a large population of samples when the prevalence rate of an outbreak is low in the geographical area. Laboratories could potentially benefit from sample pooling by increasing its testing capacity, reducing the staff number, reducing reporting time, and reducing cost. With over 20 years of experience in the testing service industry, HKLife has accumulated valuable knowledge that we are happy to share with our customers and partners. We have previous experience in helping other companies to establish new laboratories for new testing services and we are glad to help you too. If you need any help, please feel free to contact us.

Make sure your qPCR system has been professionally calibrated. Fluorescence reader without proper calibration could result in increased noise during lag phrase. This would greatly affect the threshold and Ct value determination. If you’re still experiencing problems, our team of scientists would like to have more information to help you tackle the problem. Please contact us.

Yes. Our detection kits are compatible with any extraction kit that extracts high quality nucleic acid from clinical samples. Clinical samples include nasopharyngeal swabs, oropharyngeal swabs and saliva.

The C/Tech SARS-CoV-2 RT-PCR kit requires instruments that have the FAM, HEX (or VIC) and Cy5 fluorescence channels. The C/Tech SARS-CoV-2 and Influenza+ kit requires instruments that have the FAM, HEX (or VIC), Texas Red and Cy5 fluorescence channels.

The usual lead time is 7 days for the dispatch of C/Tech SARS-CoV-2 testing products. However, due to recent outbreak and increased demand. The lead time might be slightly postponed. Please contact us for precise delivery schedule.

HKLife accepts international orders and ship internationally. We use courier service for our product shipping. Shipping time is approximately 4 days including custom clearance. If you are interested in our product, please contact us for more information.

Deltacron is supposedly a variant that has mutations that are characteristic of both delta and omicron, so is a combination of both. But there is little evidence for this variant as yet.
